Use of carbacycline derivs. - for treating atopic disorders, e.g. dermatitis, allergic rhinitis and bronchial asthma

Carbacylin derivs. (I) can be used in the prepn. of an agent for the treatment of atopic disorders. USE/ADVANTAGE - (I) can now be used to treat atopic dermatitis, allergic rhinitis and bronchiral asthma. (I) inhibit TNF synthesis at the stage of TNF-messenger-RNA. (I) are used in preference to monoclonal antibodies (targetted against TNF) in that they are effective against TNF already secreted and can also act prophylactically and in contrast to monoclonal antibodies can be used topically. Also, the TNF-TNF-immune complex formed is not degraded. Suitable doses for topical use are 20-40 micro g/g/ointment/100 cm3 affected skin; the max tolerated daily dose is 80-160 mg. Nasal sprays are used in doses of 0.1-1 ng/kg/min.
