<p>A method of producing taxanes, including taxol, in cell culture is disclosed. The method includes forming Taxane Synthesis Committed cell population aggregates which contain proliferating meristematic cells with relatively large nuclei and abundant and dense cytoplasmic contents. The aggregates are cultured, in the presence of auxin and cytokinin, and preferably in the absence of casein hydrolysate and light, and on a solid, semi-solid or layered media, to produce an increase in the size and number of plastids in the proliferating cells, with localization of taxanes in the plastids. Taxanes are obtained by disruption of the cells at harvesting. Also disclosed are methods of producing, handling and maintaining the TSC cell population aggregates.</p> |
