<p>PURPOSE:To obtain the subject substance useful as a fluorescent paint, a reagent for fluorescent antibody technique, etc., by culturing strawberry cell on a solid medium incorporated with auxin, etc., culturing the produced callus in a liquid medium incorporated with auxin, etc., and extracting a fluorescent substance from the cultured cell. CONSTITUTION:The objective fluorescent substance has high stability and emits intense green fluorescence of about 480nm wavelength by excitation with a long wavelength ultraviolet radiation having a wavelength of about 340nm. The substance can be produced in large quantities at an arbitrary time by culturing a strawberry cell on a solid medium incorporated with an auxin (e.g. 2,4-dichlorophenoxyacetic acid) or an auxin and a cytokinin (e.g. benzyladenine), culturing the obtained callus in a liquid medium incorporated with an auxin or an auxin and 2 cytokinin, extracting a fluorescent substance from the cultured cell with hot water and fractionating and purifying the extracted liquid by ion-exchange chromatography.</p> |
