PEPTIDE IN MAIN ALLERGEN DER-F II OF ACARID

PURPOSE:To obtain the subject compound for prevention, treatment and diagnosis of various allergic diseases having a specific amino acid sequence and having a property capable of binding to human anti-Der-fII IgE antibody against main allergen of Dermatophagoides, and capable of mass-producing by gene engineering. CONSTITUTION:A plasmid retaining cDNA of Der-fII as main allergen of Dermatophagoides is completely digested with a restriction enzyme KpnI in a synthetic nucleotide adaptor and subjected to 0.7% agarose electrophoresis to separate the digested DNA fragment. The DNA fragment completely containing Der-fIIc DNA is extracted and purified from agarose gel after the electrophoresis the projected terminal of cut site is smoothed and then bound to a manifestation vector to prepare the recombinant plasmid and the plasmid is inserted into Escherichia coli to transform the plasmid. The transformant is cultured in a culture medium to provide the objective peptide containing an amino acid sequence expressed by the formula, necessary to bind human anti-Der-f IgE against Dermatophagoides main allergen (Der-fII).
