PURPOSE:To provide a new lipase produced by Pseudomonas cepacia S16-019B strain, showing a high decomposition activity of vegetable and animal fats and oils at reaction conditions of a high temperature and a weak alkali and useful, e.g. as an admixture for a dish washer detergent. CONSTITUTION:Pseudomonas cepacia S16-019B strain (Bikoken stipulation No. 12574) is inoculated in a liquid medium and shaking culture is carried out for 67hr. The culture solution is subsequently centrifuged to remove the cell bodies and the resultant supernatant is collected. To the collected supernatant, solid ammonium sulfate is added so as to be 80% saturation followed by centrifugation. The resultant precipitate fraction is collected dissolved in a buffer solution and, purified by an anion-exchange column chromatography, thus giving the objective new lipase showing a high decomposition activity of vegetable and animal fats and oils at conditions of a high temperature and a weak alkali, showing a high activity in a nonionic surfactant or a nonionic surfactant-containing detergent, having 4-9.5 stable pH range, 5-6.5 optimum pH and 50-70 deg.C active temperature at pH 5.6, resistive to treatment at 50 deg.C for 2hr or at 60 deg.C for 1hr without deactivation, capable of liberating fatty acids at 1 and 3 positions of triglyceride and having 36000 molecular weight. |
