To prepare a biosensor comprising an insulating support 1 provided with a conductive film 4 acting as electrodes, on which a layer 5 is arranged comprising active molecules A, for example glucose peroxidase, a monomolecular layer of amphiphilic molecules B coupled with the molecules A is deposited on the support provided with the conductive film 4. The Langmuir Blodgett method is employed for this purpose with a bath containing the molecules A and a compression of the monomolecular layer formed on the bath is carried out, with a view to its deposition on the support, with a high surface tension P, for example of 45 mN/m. This makes it possible to obtain a compact organisation of the molecules A and to make them more accessible to the compound to be detected. The performance of the biosensor can be improved further by carrying out a surface treatment of the film 4 before the deposition of the layer 5. <IMAGE>
