<p>Liposome-based immunoassay method is carried out by coupling a liposome incorporating a marker with antibody 1 for the target antigen; reacting the liposome-coupled antibody I with antibody 2 for the target antigen and with the target antigen itself; allowing the liposome to release the marker in the presence of a complement in proportion of the amt. of target antigen present; measuring the released marker; using complement-activating antibody 3 which binds to the antibody 2 directly or indirectly (e.g. to a hapten attached to antibody 2). The method may be carried out using a kit comprising (1) the liposome-attached antibody 1; the antibody 2; the complement-activating antibody 3; the complement. The antibody 3 is pref. of horse or mouse monoclonal origin. The hapten attached to antibody 2 is pref. a low-mol.wt. gp e.g. 2,4-di, 2,4,6-tri-nitrophenyl, biotinoyl, iminobiotinoyl, or a residue of aldosterone, 17-beta-estradione, testosterone, diphenylhydantoin or isothiocyanatofluorescein. The marker is pref. carboxyfluorescein, sulphorhodanine, HRP enzyme, lucigenin etc.</p> |
