<p>PURPOSE:To provide a new physiologically active peptide inhibiting the activity of blood coagulation factor VIII, free from side effect such as hemorrhage and inhibitory action to blood coagulation factor V and useful as a therapeutic agent, etc., for blood coagulation system abnormality diseases such as thrombosis. CONSTITUTION:A peptide chain is synthesized by successively bonding amino acids having t-butyloxycarbonylamino-protecting group to a solid phase using 4-oxymethyl-phenylacetamide methyl carrier by solid-phase peptide-synthesis process. After completing the synthesis, the peptide is separated from the carrier and purified by reverse-phase high-performance liquid chromatography to obtain the objective peptide active to inhibit the activity of blood coagulation factor VIII and free from inhibitory action to blood coagulation factor V. The function of the peptide can effectively be exhibited by bonding the peptide to a protein such as bovine serum albumin, ovalbumin and keyhole limpet hemocyanine in the presence of a cross-linking reagent.</p> |
