PRODUCTION OF RECOMBINANT LIPASE

PURPOSE:To produce the subject enzyme by using a replicon of a Gram-negative bacterium wide host vector capable of replicating Pseudomonas and a recombinant capable of manifesting lipase gene by a tac promoter to make mass-production of the lipase possible. CONSTITUTION:A replicon of a Gram-negative bacterium wide host vector, capable of stably retaining in a Pseudomonas bacterium as the host and replicable is treated with a restriction enzyme and the resultant treated replicon is bound to a coil bacillus-derived tac promoter capable of manifesting its functions in a Pseudomonas bacterium. A lipase gene cloned from Pseudomonas fragi IFO 12049 strain is bound to the down stream of the tac promoter to obtain a recombinant plasmid capable of manifesting the lipase gene in the Pseudomonas bacterium. The obtained recombinent plasmid is introduced into the Pseudomonas bacterium for transformation and the resultant transformat is subsequently cultured, thus mass-producing the objective recombinent lipase.
