The base sequence of large single stranded nucleic acids is determined by retarding the migration rate of a nucleic acid fragment of n &dagger& 1 bases below a migration rate which would otherwise be the same as the migration rate of a nucleic acid fragment of n bases in a polyacrylamide gel under continuous field gel electrophoresis.
