Living vaccines for immunisation of animals against rabies contains a variant of SAD (Street Alabama Dufferin) virus which retains its activity, in liq. form, for at least 3 days at at least 20 deg.C. Pref. the variant is VA1, VA12, VA17 or VA20. It is apathogenic in adult mice when administered intracerebrally and remains so after intercerebral passages in baby mice. The variant VA1 (C.N.C.M. I-430) is isolated from the BSR-19 strain by cloning on soft agar. From the variant VA12 (I-431) or VA17 (I-432) are recovered using the monoclonal antibody MW-187.2.8, while VA20 is isolated from VA12 using the monoclonal antibody PM13. The variant is grown on the cell line BMK-BSR in monolayers in several stages at split rate pref. 1:3-10. Every 2-5 days virus is harvested and stored at -20 to -80 deg.C. The resulting virus suspension is made into a vaccine with e.g. pH 7.2 buffer; bovine serum albumin and ovalbumin, to a virus concn. 30-40 million 50% infectious doses per inoculation (1.8ml).
