Method of determining an amino acid sequence of a protein or a peptide, wherein the protein or peptide is sequentially degraded from its C-terminal to give a reaction mixture, by treating with (a) a vapour containing an organic acid, or (b) an anhydride of an organic acid, where the organic acid is CF3 -(CF2)n - COOH, where n is zero or an integer. The reaction mixture is then analysed to determine the amino acid sequence of the protein or peptide, for example using a mass spectrometer or an amino acid analyser. |
