PURPOSE:To read from a separating area of a large fragment of nucleic acid to a fluorescent pattern of a minute part to determine the sequence of each base in a series of operations by selecting a dyeing agent and a labelling fluorescent substance and, using a common nucleic acid electrophoretic pattern reading apparatus. CONSTITUTION:A sample fragment of nucleic acid is supplied from above the gel in an electrophoretic part 5 to perform electrophoresis after an electrophoretic voltage is applied. The gel after completing the electrophoresis is set in a main body 7 of a measuring part. The electrophoretic pattern of the gel is detected and made visual through a processor 8 and a printer 9. At this time, tire sample nucleic acid is separated by the electrophoresis of the gel, and dyed with ethidium bromide so as to confirm the position of the band of nucleic acid. Then, the electrophoretic pattern of the sample is excited with a laser light including 532nm wavelength, and the generated pattern of the distribution of the fluorescence intensity is read in the wavelength region of approximately 600nm, thereby to select the determining part of the sequence of the base of the sample. Further, Rhodamine X is selected as an example of a labelling fluorescent substance to label the sample. As the labelled sample is subjected to electrophoresis, the fluorescence pattern is read. |
