PROCESS FOR THE PREPARING APROTININE ()

The method is applicable in the pharmaceutical industry for preparation of aprotinin, which acts as a trypsin and kallikrein inhibitor. This is a simplified method, which enhances the yield and efficiency of raw aprotinin purification. According to the method, 70 per cent ethanol-water extracts from bovine lungs or bovine external ear glands with a pH of 3.5-4.0 are adsorbed in a column filled with a polystyrene-based macroporous sulphocationite, meshed with divynilbenzole. The ballast substances are desorbed in a step-wise fashion from the ionite with eluents with a pH of 8.5-9.5, containing up to 0.3M sodium chloride. The raw aprotinin is desorbed with eluents with a pH of 9.5-11.0, containing up to 1.0M sodium chloride, for 2-3 hours and is precipitated with 3-5 volumes of cooled acetone. Then follow gelchromatographic separation and lyophilization.
