PURPOSE:To obtain a new recombinant microorganism useful for secreting and producing a large amount of human.lysozyme. CONSTITUTION:A microorganism, e.g. Saccharomyces.cerevisiae E91 strain (FERM P-1432) which is a recombinant microorganism, holding a plasmid containing human.lysozyme gene, having the ability to highly secrete the human.lysozyme and belonging to the venus Saccharomyces. The aforementioned microorganism is obtained by preculturing a transformant Saccharomyces.cerevisiae (KK4 (PESH)) of Saccharomyces.cerevisiae KK4 strain with a plasmid vector PESH having ENO1 promoter and the human.lysozyme gene bound to a secretory signal in a synthetic liquid culture medium without containing leucine, then subjecting the resultant culture to mutation treatment with ethylmethanesulfonic acid, diluting microbial cells, subsequently smearing the diluted microbial cells on an agar plate for sorting out a mutant strain capable of highly secreting the human.lysozyme and sorting out 4 strains (E74, E82, E91 and E95) forming especially large-sized halos. |
