Selective quantitative determination of individual lectins in plant preparations (A) contg. a mixt. of lectins comprises, in an ELISA procedure, (1) reacting (A) with carrier-bound neoglycoprotein (NGP); (2) treating NGP-bound lectin with a blood serum contg. lectin antibodies (Ab); (3) treating lectin-bound Ab with a labelled anti-antibody (AAb) and (4) determining the amt. of label bound to provide a value of the specific lectin concn. (by comparison with predetermined standard values). Specifically, NGP is chosen to provide the required binding activity and selectivity for a particular lectin; these properties are determined by inhibition tests in presence of sugar or sugar derivs. with one competitive with regard to the sugar component of NGP. USE/ADVANTAGE - To quantify the mistletoe (Viscum album) lectins MLI and MLII (active ingredients in mistletoe extracts used for cancer treatment). It provides reliable results even in very dilute preparations and can differentiate between closely similar lectins.
