PURPOSE:To obtain the subject new polypeptide useful for diagnosing hepatitis C and testing, etc., blood for transfusion by fusing a hepatitis C-related antigen- derived antigenic polypeptide to a polypeptide of a kind different from that of the aforementioned antigenic polypeptide through methionine. CONSTITUTION:A DNA capable of coding hepatitis C-related antigenic polypeptide derived from hepatitis C viruses(HCV) is synthesized by using a solid-phase synthetic method according to a phosphoramidite method, then treated with a restriction enzyme and linked to a linker for linking to a DNA capable of coding a polypeptide of a kind different from that of the above-mentioned antigenic polypeptide. The resultant linked DNA is subsequently linked to a plasmid vector and a promoter, etc. A DNA capable of coding chum salmon growth hormone is synthesized as a polypeptide of a kind different from that of the aforementioned antigenic polypeptide, treated with a restriction enzyme and integrated into the above-mentioned plasmid to prepare a DNA capable of coding a fused antigenic polypeptide in which both the polypeptides are linked through methionine. The obtained DNA is then transduced into a microorganism and expressed to afford the objective fused antigenic polypeptide. |
