The invention relates to a process for cloning nucleic acids, comprising a step for ligating, to the 3' end of a primer extension product, a single-stranded oligonucleotide, non-complementary to the said primer extension product, and a step for synthesising the second strand using a primer complementary to the said oligonucleotide, optionally followed by amplification reactions.
