An efficient method for detecting loose smut infection in the embryos of barley and wheat seeds utilizes different concentrations of sodium hydroxide and granular sodium chloride to separate the embryos from the husks and endosperms. The floating embryos are skimmed off with a strainer. Then they are cleared in lactic acid, and the mycelium stained with trypan blue. Infected embryos can be observed with the aid of a stereoscope at 60x magnification. |
