PURPOSE:To prepare a large amount of tryptophan composed exclusvely of L- isomer, by the use of microorganisms capable of producing the objective compound from indole and serine, or from indole, pyruvic acid and ammonium ion. CONSTITUTION:Caulobacter crescentes ATCC19089, etc. belonging to Caulobacter genus are cultured in a conventional synthetic or natural medium containing triptophan. The cultured product or its treated product is suspended in a reaction liquid containing a substrate composed of indole and serine, or indole, pyruvic acid and ammonium ion, and subjected to the enzymatic reaction. The reaction liquid pref. contains ethylenediaminetetraacetic acid and pyridoxal phosphate. The produced L-triptophan can be separated by conventional ion exchange resin treatment, etc. |
