摘要 |
<p>PURPOSE:To select a pigment producing cell in high efficiency by dyeing a pigment producing cell with a fluorescent reagent emitting fluorescence in the same waveband as that of the absorption wavelength of the pigment and separating a cell having the fluorescence neutralized with the pigment by a flow cytometer. CONSTITUTION:A pigment producing cell (e.g. anthocyanin highly producing cell) is put in a solution of a mixture of cellulase with pectinase and incubated to isolate a protoplast. The protoplast is dyed with a fluorescent reagent (e.g. fluorescein isothiocyanate) of emitting fluorescence in the same waveband as that of the absorption wavelength of the pigment produced, a cell having the fluorescence of the fluorescent reagent neutralized with the pigment is separated by a flow cytometer having sorting function to select a pigment producing cell having high productivity of reddish pigment. A fluorescent reagent having an excitation waveband excitable with light rays at >=454.5nm wavelength is preferably used as the fluorescent reagent.</p> |
