<p>PURPOSE:To efficiently obtain the title antibody by reacting a hybridoma prepared by cell fusion with an antigen immobilized carrier, separating the reacted hybridoma to form a monoclonal antibody and facilitating screening. CONSTITUTION:A protein antigen is blended with a Freund's complete adjuvant, administered to an abdominal cavity of BALB/c mouse, the mouse is immunized and the similar immunization is continued at intervals of several weeks. After the final immunization, splenocytes are collected, subjected to cell fusion with a myeloma cell line by using polyethylene glycol and selectively cultured by using a HAT medium to give a hybridoma. The hybridoma obtained by the cell fusion is brought into contact and reacted with a carrier to which the protein antigen initially used for immunization is immobiliged, the medium is eliminated, the carrier is separated, cleaned, cultured, a positive clone bound to the carrier is multiplied, cloned by limiting dilution method and cultured to give the objective monoclonal antibody.</p> |
