Preparing antibodies against unstable antigens, esp. active anaphylatoxin - by direct contact of immunogen and spleen cells, useful diagnostically and for treating elevated complement activation

Antibodies (Ab) are produced by directly contacting splean cells with at least one unstable immunising antigen (Ag). Also new are (1) anti-anaphylatoxin antibodies (Ab1), specific for the active form of anaphylatoxin (I) and prepd. by this method and (2) cell lines able to produce Ab1. More specifically, Ag is a material which undergoes rapid enxymatic metabolism in the body of the animal being used for Ab prodn.. Partic. it is a protein and/or peptide, specifically the active forms of anaphylatoxins C3a,C4a and/or C5a. USE/ADVANTAGE - Ab1 are used (1) therapeutically in cases of enhanced complement activation (esp. admin. by intravenous injection); or (2) diagnostically for quantitative determn. of the active forms of (I). Ab1 do not react with either inactive (I) or native complement factors.
