PURPOSE:To specifically and sensitively detect nucleic acid in a short time by using plural oligonucleotide as a primer for a chain-lengthening reaction, enzymatically amplifying a specified DNA contained in the target DNA, subsequently adding another primer, carrying out amplification and acting a stain thereon. CONSTITUTION:The first reaction for selective amplification of a specified DNA contained in the target DNA by a DNA synthetase is carried out by making two kinds of oligonucleotides function as a primer for a chain- lengthening reaction. The primer used in the first reaction and one kind or two different kinds of primers are then acted on the resultant amplified DNA to carry out the second reaction for further selective amplification of the DNA fragment in this DNA by a DNA synthetase. After completion of the reaction dyeing is carried out by acting a stain thereon and the absorbance or the fluorescence intensity of the above-obtained dyed solution is then measured to detect the amplified DNA fragment. The target DNA is highly specifically and sensitively detected in a short time thereby. |
