PURPOSE:To obtain a surface DNA useful for production of treating agent for immunodeficiency or tuner by isolating a clones DNA coding human interleukin 1 precursor polypeptide using a human HL-60 cell and then subjecting the isolated cloned DNA to restriction enzyme treatment. CONSTITUTION:A human acute myeloid leukemia strain cell (human HL-60 cell) is cultured in a culture medium containing a differentiation inducing agent and a cell differentiated from the cultured mixture is collected and treated with a inducing agent such as enditoxin and then RNA is extracted by an ordinary method and subjected to oligo(dT)cellulose column to separate mRNA. Cyclic DNA is synthesized using this mRNA as a template and the cDNA is subjected to restriction enzyme treatment, bound to a vector and introduced into a host to prepare cDNA library, which is then cultured and is subjected to cloning using a gene fragment coding interleukin 1 as a probe to select positive clone. A plasmid is extracted from the clone and then subjected to restriction enzyme treatment to produce DNA coding a human interleukin 1alpha polypeptide expressed by the formula.
