<p>PURPOSE:To readily and rapidly produce the subject compound with high condensation degree by deprotecting 5' position of ribonucleotide in which 2' and 5' positions are protected by a tetrahydropyranyl group and a 9-phenylxanthenyl group, respectively, and then bonding a similar protected derivative thereto. CONSTITUTION:Ribonucleotide in which 2' and 5' positions are protected by a tetrahydropyranyl group and a 9-phenylxanthenyl group, respectively, is treated with 1.5-2.5vol dichloroacetic acid-containing organic solvent solution to eliminate the protecting group at 5' position. The resultant compound represented by formula I (R' is 9-phenylxanthenyl, etc.; R''' is substituent group; B is nucleic acid base) is condensated with a phosphoroamidite unit-containing ribonucleotide derivative of formula II (R is tetrahydropyranyl; R'' is protecting group), etc., in which 2' and 5' positions are protected by a tetrahydropyranyl group and a 9-phenylxanthenyl derivative, respectively to obtain a compound of formula III. The abovementioned reaction is repeated, thus obtaining the objective oligonucleotide.</p> |
