PURPOSE:To obtain the title high-purity inhibitor in an industrially advantageous way by collecting from cereals, through ultrafiltration or isoelectric point fractionation, an inhibitor specifically acting on the lipase originating from the human or animal stomach or pancreas. CONSTITUTION:Cereals (e.g. wheat) are immersed overnight at room temperature in a 0.1M tris hydrochloric acid buffer solution (pH T.2) containing 10% saturated ammonium sulfate and 50mM calcium acetate and then ground, and the resulting extract liquor is centrifugalized to collect a supernatant, which is, in turn, subjected to a ultrafiltration module with a fractionating molecular weight of 150Kd, and the filtrate is collected and incorporated with a 0.5-1.0M neutral salt such as NaCl to remove enzymes (e.g. amylase) bound to lipase inhibitors followed by fractionation using a second ultrafiltration module 50-70Kd in fractionating molecular weight, and fractions with molecular weights of 50-70Kd or higher are collected and then made into powder through e.g. lyophilization, thus obtaining the objective lipase inhibitor specifically acting on the lipase originating from the human or animal stomach or pancreas. |
