<p>PURPOSE:To obtain the subject monoclonal antibody, capable of specifically recognizing a human gliacyte growth inhibiting factor, inhibiting the activity of the human gliacyte growth inhibiting factor and useful for purifying and determining the human gliacyte growth inhibiting factor and treating Alzheimer's diseases, etc. CONSTITUTION:Tumorous masses of neurofibroma obtained from a patient suffering from Recklinghausen's disease are homogenized in a buffer solution and subsequently centrifuged to collect a supernatant, which is then successively purified by a gel filtration column, an anion exchange column, hydrophobic chromatographic column and a hydroxyapatite column to isolate a human gliacyte growth inhibiting factor (human GGIF). The resultant factor is used as an antigen, emulsified and administered to immunize BALB/C female mouse. After the final immunization, splenic cells are collected and fused to mouse myelomatous cells. The obtained fused cells are cultured to select a positive clone capable of producing an antibody against the human GGIF. The selected clone is cloned and subsequently cultured to afford the objective human monoclonal antibody capable of specifically recognizing the human GGIF.</p> |
