PURPOSE:To profitably produce amylase playing an important role in a starch- saccharifying industry by culturing a novel amylase-producing bacterium belonging to the genus Fusidium and subsequently collecting the product from the cultured product. CONSTITUTION:The production of amylase in a high yield comprises following processes; culturing a novel amylase-producing bacterium [e.g. Fusidum.SP-BX-1 (FERM 12164)] belonging to the genus Fusidium in a medium containing 0.5% of yeast extract as a nitrogen source and a carbon source such as glycerol or starch at 30 deg.C for three days under rotation and vibration, centrifuging the cultured suspension to separate bacterial cells, filtering the supernatant, adding ammonium sulfate to the filtrate, recovering a fraction precipitated within a range of 30-75% saturation, dissolving the fraction in a buffer solution and subsequently applying the solution to an ion exchange column and to a gel filtration in turn for the purification of the amylase. |
