PRODUCTION OF HYBRIDOMA

PURPOSE:To obtain the subject cell capable of producing a monoclonal antibody of the IgG class against an antigen which is difficult to produce the IgG by culturing a hybridoma capable of producing a monoclonal antibody of the IgM class together with its antigen and a thymocyte. CONSTITUTION:A hybridoma (e.g. a rat hybridoma KM-602) capable of producing a monoclonal antibody of the IgM class reactive with a selective ganglioside GM2 of the hybridoma, together with an antigen reactive with the aforementioned antibody and a thymocyte (e.g. a thymocyte derived from a mouse), is added to a culture solution and cultured while carrying out stimulation of the antigen and subjected to class switch. Thereby, the objective hybridoma capable of producing the monoclonal antibody belonging to the IgG class is obtained. Furthermore, the aforementioned hybridoma capable of producing the monoclonal antibody of the IgG class is cultured in a culture medium by using a roller bottle, a spinner, etc., for 2-7 days, then centrifuged, passed through a protein A column and purified to afford the objective monoclonal antibody of the IgG class.
