<p>PURPOSE:To provide an anti-idiotype monoclonal antibody specifically reactive with anti-sialyl Lewis A monoclonal antibody, having a structure similar to antigen epitope of sialyl Lewis A and utilizable as a diagnostic for cancer, a cancer vaccine, etc. CONSTITUTION:A spleen cell of an animal immunized with a complex of an anti-sialyl Lewis A monoclonal antibody and a carrier protein is fused with a mouse myeloma cell to form a hybridoma. A hybridoma (FERM BP-3334) capable of producing monoclonal antibody inhibiting the reaction of sialyl Lewis A antibody and anti-sialyl Lewis A monoclonal antibody is selected from the above hybridoma and cultured in a medium or administered to an animal to affect the animal with ascites carcinoma. The cultured liquid or the ascites is collected and an antibody reactive with anti-sialyl Lewis A monoclonal antibody, non-reactive with monoclonal antibody for the other antigens and inhibited by an oligosaccharide of an epitope of sialyl Lewis antigen is selected from the obtained monoclonal antibody to obtain the objective antibody.</p> |
