Method for determining an analyte (I) comprises reaction with two specific binding partners, one of which (A) is enzymatically -labelled and unfixed to a carrier, while the other (B) is fixed to a carrier. After the specific binding reaction has occurred, the amt. of unbound (A) is taken as a measure of (I), this amt. being determined using a signal-generating substrate for the enzyme label. The new feature is that during the binding reaction two enzyme substrates are present; one (II) is non-fixed and generates no signal, while the other (III) is fixed to a carrier and does generate a signal. The amt. and affinity of (II) are chosen so that when simultaneous incubation with both substrates is carried out, reaction of (III) is delayed, until the specific binding reaction has gone practically to completion. Also new are test carriers for this method. (B) Specifically binds to both (I) and (A), while (A) reacts with both (I) and (B). Esp. one of them is an antibody and the other an antigen.
