PURPOSE:To obtain the subject new enzyme for producing pulp, etc., by culturing microorganisms, belonging to the genus Lentinus and having the ability to produce phenol oxidase LE in a culture medium and reducing the size of lignin molecules or decomposing the lignin. CONSTITUTION:SHIITAKE mushrooms belonging to the genus Lentinus are inoculated into a liquid culture medium, cultured at 28 deg.C and then filtered with a filter cloth to collect a culture supernatant, which is then purified by gel filtration and ion exchange column chromatography. Thereby, the objective enzyme capable of catalyzing alkylphenyl cleavage and oxidation of the alpha-position of a lignin model compound, oxidizing o-phenylenediamine, producing a red pigment, oxidatively decomposing syringylglycerol-beta-guaiacyl ether, oxidizing guaiacol and catechol, etc., without oxidizing tyrosine and DOPA, having 4.5 optimum pH, 550 optimum temperature, about 3.0 isoelectric point and 65000 molecular weight and inhibited by NaN3 and Fe<2+> is obtained. |
