<p>PURPOSE:To readily detect soil pathogenic molds as a diagnostic agent for infection of plants with soil pathogenic germs of the genus Fusarium by measuring a plant tissue, etc., according to a fluorescent antibody method using a monoclonal antibody specific for the soil pathogenic germs of the genus Fusarium. CONSTITUTION:A soil pathogenic germ (e.g. Fusarium oxysporum 860926a) belonging to the genus Fusarium is cultured in a dextrose liquid culture medium and then subjected to centrifuging treatment to collect microbial cells, which are subsequently crushed in liquid nitrogen and suspended in a phosphoric acid buffer solution to provide an antigenic suspension. The resultant antigenic suspension is then administered to immunize Balb/c mouse. After the final immunization, splenic cells are collected and fused to mouse myelomatous cells and then subjected to screening and cloning to afford a hybridoma, which is subsequently cultured to provide a monoclonal antibody specifically reactive with microbial cells of the soil pathogenic germs of the genus Fusarium and a milled microbial cell liquid. The resultant monoclonal antibody is used to detect the soil pathogenic germs of the genus Fusarium from plant tissual pieces, plant juices, etc., according to a fluorescent antibody method or an enzyme immunoassay method.</p> |
