PURPOSE:To provide a new antibiotic substance RK-286D active to inhibit proten kinase C and useful as an antitumor agent and antiinflammatory agent. CONSTITUTION:An antibiotic substance RK-286D expressed by formula and having the following properties. Melting point, 225-230 deg.C (browning); molecular formula, C26H28N3O4; elemental analysis, C 70.65%, H 5.35%, N 9.61%, solubility, easily soluble in dimethyl sulfoxide, etc., and insoluble in water; molecular weight, 441.1692 (HR-EIMS); color reactions, positive to Reydn-Smith reaction, etc., and negative to ninhydrin reaction, etc.; color, pale yellow. The antibiotic substance can be separated from the culture product of an RK-286D producing microorganism of genus Streptomyces, e.g. Streptomyces sp. RK-286 (FERM P-10634). The culture is carried out under nearly neutral pH condition at 25-35 deg.C preferebly by aeration and agitation process. |
