A human insulin-like growth factor I (IGF-I) is prepd. by (a) treating pYLAC-A/G-SOD with Pst I, Sal I, Nco I and Pst I to obtain the gene fragment contg. termination gene sequence (I) of glucoaldehyde-3-phosphate dehydrogenase (GAPDH), fragment contg. mixed operating gene sequence (II) of alcohol dehydrogenase/glucoaldehyde-3-phosphate dehydrogenase (ADH/GAPDH), (b) treating M13mp18 RF vector with XbaI and Bsph I to obtain the leader sequence gene fragment (III), (c) treating M13mp18-IGF-I with Xba I and Sal I to obtain human IGF-I gene fragment (IV), and (d) ligating (I), (II), (III) and (V). The obtd. human IGF-I expression vector, pYLBC-A/G- F-IGF-I, is composed of mixed operating gene (ADH-2/GAPDH), alpha factor leader sequence, human IGF-I gene and GAPDH termination gene.
