PURPOSE:To obtain a new secretory vector of producing a large amount of hirudin of an anti-blood coagulation factor, containing a DNA sequence encoding replication starting site of PUC plasmid, Tac promoter, etc., signal peptide and foreign protein, respectively. CONSTITUTION:Escherichia coil is transformed by using a secretory vector containing a DNA sequence obtained by cleaving plasmid pUC18 with restriction enzyme PvuI and PvuII, as a DNA sequence containing replication starting site (ori) of PUC plasmid, a DNA sequence of TAC promoter of Trp promoter, a DNA sequence encoding a signal peptide derived from an alkali phosphatase and DNA sequence encoding a heterogeneous protein such as a hirudin variant having an amino acid sequence shown by the formula. The transformed bacilli are cultured, products are recovered from the medium to produce a large amount of heterogeneous proteins such as hirudin and isomers thereof. |
