<p>PURPOSE:To provide the subject derivatives useful as an intermediate for synthesis of isoprenoid type phospholipids capable of producing a membrane, excellent in biocompatibility and membrane flow properties with high barrier properties, and also excellent in dispersion stability and chemical stability and capable of forming an intermolecule association. CONSTITUTION:A natural diterpene, (7R,11R)-phytol of formula I is hydrogenated in the presence of platinum oxide catalyst, subsequently oxidized in the presence of ruthenium trichloride catalyst to obtain a carboxylic acid derivative. The resultant carboxylic acid derivative is then converted to its acid chloride using thionyl chloride and then made to react with 3-benzyl-sn-glycerol in the presence of diisopropylethylamine. Benzyl group is subsequently eliminated to obtain a glycerol-1,2-diester derivative represented by formula II. This derivative is then allowed to react with diphenylphosphorochloridate in the presence of diisopropylethylamine to obtain its triphosphate and the resultant triphosphate is subsequently reacted with hydrogen in the presence of platinum oxide, thus obtaining the objective glycerol derivative shown by formula III (R and R' are H or phosphoric acid-protecting group; (n) is 1-3).</p> |
