The present invention is directed on a composition of thermophilic enzymes as a tool for DNA synthesis (in vitro), the composition comprising, in combination, an enzyme being capable of carrying out the polymerization of dNTP to form (de nova) synthesized DNA molecules within the temperature interval of 56-90 C (thermophilic DNA-polymerase) and an enzyme being capable of eliminating accumulated pyrophosphate (thermophilic pyrophosphatase) within the same temperature interval. The invention is also directed on the preparation and the use of the said composition.
