PURPOSE:To stably industrially produce a new plasmid by culturing a microorganism belonging to the genus Micromonospora and subsequently extracting the cultured product. CONSTITUTION:Micromonospora . griseorubida A11725 strain is cultured in a medium containing glucose, etc. The cultured product is centrifuged, and the obtained cells of the microorganisms are extracted to provide a plasmid having a mol. wt. of 18.6-19.6 mega dalton, and having a scission map by various restriction enzymes in which sensitive sites numbers against enzymes are 6 by BamHI, 4 by Bgl11, 6 by Bcl1, 2 by ClaI and 4 by Kpn1. If necessary, the plasmid is combined with a desired vector marker by the utilization of the autonomous replication ability thereof to provide a cloning vector containing a DNA fragment having a mol. wt. of 17.4 0.5mega dalton and containing a drug-resistant gene, and having sites sensitive against various restriction enzymes at BamHI 6, Bgl11 2, Bcl1 5, ClgI 3, and Kpnl 3. |
