PURPOSE:To easily extract an enzyme produced by a Gram-negative bacteria from the bacteria on an industrial scale by contacting glycine to the culture system during and/or after the cultivation of Gram-negative bacteria. CONSTITUTION:Enzyme produced by a Gram-negative bacteria is separated and eluted from the bacterial cell by contacting the cell with glycine during and/or after the cultivation. The enzyme produced by the Gram-negative bacteria is e.g. tryptophan synthetase (A) and tryptophanase (B). The bacterial strain capable of producing the enzyme A is e.g. Escherichia coli and Pseudomonas putida, preferably Escherichia coli. The strain capable of producing the enzyme B is e.g. Escherichia coli and Proteus rettgeri, preferably Escherichia coli. The amount of glycine to be added to the system during or after the cultivation is usually about 0.01-30%(w/v). |
