PURPOSE:To obtain an improve carrier for immobilizing enzyme, by treating porous, granular chitosan with an excess amount of an active ester of a dicarboxylic acid so that the chitosan contains an reacted active ester and reacting the active ester with an amino group of an enzyme. CONSTITUTION:Low-molecular weight chitosan is dissolved in an aqueous solution of acetic acid, etc., and dripped in a coagulating bath to give porous, granular chitosan. The porous, granular chitosan is stirred with an excess amount of an active ester of dicarboxylic acid in a solvent and crosslinked. A dicarboxylic acid di-p-nitrophenyl ester, dicarboxylic acid di-N-hydroxy- succinimide ester, etc., are used as the active ester of the dicarboxylic acid. After the reaction, the solvent is removed to give granular chitosan for immobilizing enzyme. The carrier contains an unreacted active ester, is readily reacted with an amino group of an enzyme in water under a mild condition and the enzyme is easily immobilized. |
