The invention comprises an antibiotic 19402 R.P., active against gram-positive micro-organisms, which is an acid forming salts with the alkali metals, the sodium salt of which has the following characteristics:-a white amorphous powder which is soluble in water, soluble in anhydrous methanol, and in ethanol, propanol and isopropanol containing water, very sparingly soluble in anhydrous ethanol, propanol, isopropanol and butanol, and insoluble in acetone, hexane, benzene, diethyl ether, chloroform and ethyl acetate; elementary analysis C=46À25%, H=6À9%, 0=36À7%, N=4À1%, P=2À4%, S=0À6%, Na=3À05%; U.V. spectrum max. at 256 nm (E<SP>1%</SP> 1cm. = 102À5), min. at 227 nm (E<SP>1%</SP> 1cm. = 18À7), I.R. according to the drawing; [α]<SP>20</SP> D = 1À9Œ1‹ (C=0À4% water); and Rf values according to Table II (not shown). The antibiotic is produced by cultivating Streptomyces peruviensis NRRL 2757 or a 19,402 R.P. producing mutant thereof, in an aqueous nutrient medium. It is isolated from the fermentation broth by adjusting the pH of the medium to below 5À0, filtering, extracting the filter cake with water containing a C 1-5 aliphatic alcohol at pH 3 to 7, concentrating in vacuo below 40‹ C. and precipitating the antibiotic by the addition of a non-solvent, e.g. a mixture of ethanol and acetone. If the pH is adjusted to 7À0 with an alkali metal hydroxide, the crude antibiotic salt will precipitate. Further purification is by (A) adsorption on an anionic ion-exchange resin followed by elution with aqueous alcohol containing an electrolyte, followed by dialysis to remove the salts; (B) chromatography on silica gel followed by elution with a mixture of n-propanol and ammonia. A pharmaceutical composition comprises the antibiotic 19402RP or an alkali metal salt thereof and a carrier. |
