PURPOSE:To effectively break liposome without inactivating a released liposome content by subjecting the liposome containing phospholipid as the structural component to enzyme treatment with phospholipase. CONSTITUTION:Liposome containing phospholipid as the structural component is broken by enzyme treatment using phospholipase such as phospholipase A2. This enzyme hydrolyzes acylester coupled with the 2-position of glycerol and cuts the interaction of hydrophobic coupling with aliphatic chain of other phospholipid to break the molecular structure. Thereby, the sealed enzyme and labeled compd. contained in the liposome to be released after breaking can not be inactivated, and the released liposome maintains the transparency as a liquid state and gives clear color proportional to time due to the reaction of the released enzyme. |
