NEW MATERIAL:Yeast protein disulfide isomerase. Action: Isomerization catalysis of disulfide bond; Molecular weight: 70000 (Na Dod SO4-polyacrylamide gel electrophoresis); Isoelectric point: 4.1; Molecular weight after endoglycosidase H treatment: 63000; Optimum pH for disulfide isomerization activity: 8.75. USE:Utilized, e.g. for carrying out disulfide isomerization of a protein having impertinent disulfide bonds and changing the impertinent disulfide bonds to pertinent disulfide bonds so that the protein may have an active conformation. Useful also as an antigen, etc., for production of an antibody. PREPARATION:Cells of a yeast (e.g. Saccharomyces cerevisiae AH 22 strain) are initially crushed and centrifuged followed by alkaline extraction of the resultant precipitation. The resultant extract is then purified using the ammonium sulfate fractionation method in combination with the gel permeation chromatography method, the gel permeation HPLC method, etc. |
