<p>PURPOSE:To suppress germination and rooting of lump of spherical material and to form and to propagate only lump of spherical material in tissue culture of fern by controlling an amount of hormone agent to a liquid medium and carrying out a roller tube culture. CONSTITUTION:By tissue culture of fern, the tissue is inoculated to a medium and cultured to give lump of spherical material, which is separated into four to six parts, transplanted to an agar-free liquid medium obtained by adding a hormone agent to MS medium as a basic medium and subjected to roller tube culture so that germination and rooting of the lump of spherical material are suppressed and only the lump of spherical material is propagated to carry out nutritive multiplication by tissue culture of fern.</p> |
