PURPOSE:To enhance reliability in the measurement of the time required for holding a material for analysis and also optimize applied voltage by measuring the temperature of the inside of a capillary when using CZE method. CONSTITUTION:A hollow capillary 1 made of glass is filled with a phosphoric acid buffer and has a pressure reducer 2 connected to one end thereof and a small amount of a thermochromic pigment solution is injected into the capillary by means of pressure reduction. Next the content of a container 3 is changed to the phosphoric acid buffer containing e.g. serum protein transferrine and a small amount of a water solution of proteins is injected again into the capillary by means of pressure reduction. The capillary is installed in a capillary electrophoresis device. A detector 8 which measures the absorbance of the capillary at two wavelength 220 nm and 500 nm is disposed in the middle of the capillary. Light of wavelength 220 nm is used for measuring the density of the proteins and light of wavelength 500 nm is used for measuring the absorbance of the thermochromic pigment, i.e. temperature. |
