NEW MATERIAL:The non-A non-B hepatitis virus antigen polypeptide having the amino acid sequence of formula. USE:A reagent for detecting non-A non-B hepatitis virus antibody. Vaccine for preventing and curing the infection with non-A non-B hepatitis virus. PREPARATION:The objective polypeptide of formula can be produced e.g. by using the serum of a non-A non-B hepatitis patient as a starting raw material, concentrating and recovering the virus particle as a precipitate by polyethylene glycol precipitation process, extracting RNA from the virus by guanidine- thiocyanate process, etc., synthesizing a cDNA using the RNA as a template, inserting the cDNA into a phage vector to prepare a cDNA library, culturing the library, screening by enzyme immunoassay to select a positive plaque, extracting DNA from the plaque, treating with restriction enzymes, bonding to a vector, inserting into E.coli, etc., to effect the transformation of the cell and culturing the obtained transformant. |
