NEW MATERIAL:A phospholipase D having the following properties: Action, hydrolyzing the phospholipid of formula I (R1 and R2 are acyl or alkyl; X is organic group produced by removing one OH group from an OH-containing base) to isolate a phosphatidic acid and a base or converting the phosphatidyl group of a phospholipid of formula I to an alcoholic OH in the presence of a compound containing alcoholic OH such as glycerol of formula R3-OH (R3 is organic group) to form a phospholipid derivative of formula II; substrate specificity, the relative activity to lysophosphatidylcholine is 2.3 when the hydrolysis activity to phosphatidyl choline is taken as 100; optimum pH, 5.5; pH stability, stable at pH 4-8; optimum temperature, 55-65 deg.C; molecular weight, 64,000; etc. USE:Reagent for the production of phospholipid derivative and the determination of phospholipid in serum. It has high phosphatidyl group transfer activity. PREPARATION:S-170 strain (FERM P-11422) belonging to genus Streptomyces is cultured and the objective enzyme is collected from the culture product. |
