A method for producing penicillin acylase with participation of Escherich coli strain or its mutants. The method is distinctive in that the biosynthesis process is conducted with application of the new MT 40/47 mutant of Escherich coli unable to use citrate from agar substrate according to Christensen. The process is conducted in the temperature below 40 degrees C, preferably in 24 degrees C, in oxygen conditions and on simple culture media with simultaneous stirring and airing until penicillin acylase is produced in cells of micro-organisms.
